I wouldn't risk it even with a toy gun. There are plenty of replicas that say replica on the side of a firearm that have been converted to fire live ammunition.
No offense intended but if you have to ask this question you're better off just handing your valuables over. A fake just doesn't look right to someone cognizant about guns. Can you tell pot metal from real steel just by seeing it? Don't be embarrassed, most people can't tell the difference. If you're staring at the wrong end of a revolver you may (or not) have the presence of mind to see if there are any bullets in the cylinders. Of course, the bad guy could have wadcutters loaded in his revolver and you may not be able to see them. Although a wadcutter is just a target load it will inflict from serious bodily harm to life-ending injuries. For that matter a blank gun or a real gun loaded with blanks can also infict from serious to life-ending injuries.
So until you become very familiar with guns, handle them almost daily, go to gun shows and really look at what's there, go to a shooting range and practice it is very difficult for a novice to tell the difference. I'm a gun person and I've had to pickup and handle quality replica handguns and BB guns to be absolutely sure that they are not the real deal.
If you're really concerned with being robbed at gun point you really should take a gun safety course and consider obtaining a concealed weapons permit. A confident person walks with a certain air that does not target him/her as easy prey. People who resort to sticking a gun in your face to rob you are basically cowards and may augment their false-courage by shooting you anyway regardless of whether or not you comply with their demands.

Learn well how to use it, take a basic safety and a defensive shooting class.
Practice regulary.
If someone confronts you with a threat it is not your responsibility to determine the veracity of the threat only to respond appropriately to the threat.
If someone points a "gun" at you defend yourself appropriately.
A "gun" used in a malicious manner is a deadly weapon regardless of if it is capable of firing ammunition or not.
If you feel your life is threatened and your have no ability to flee then you are justified in using deadly force!
You may feel terrible if you shoot a robber using a fake gun, but remember if you are being robbed with a fake gun it is only because the robber could'nt get a real one.
His intent is still the same. To assault you using the threat of force.
If in doubt the gun is real.

To be able to tell whether a gun is real or fake requires an intimate knowledge of firearms. If i can look at the gun for a few seconds, i can usually tell if its fake or not.
that being said, it also depends on the situation. a friend trying to pull a prank on me is a completely different situation from being robbed by a stranger. Police officers who mistake toy guns for real guns usually do so because of the situation. they can't afford to guess and be wrong in a threat situation, and neither can you.
as to how to tell if a gun is loaded, it depends on the gun. if someone points a real revolver at you, you should be able to see the bullets in the end of the cylinder. there is no way to tell on a rifle, shotgun, or semi-auto pistol.

Cops frequently can't tell the difference between a real gun and an air pistol. Take a look at the airgun case at a sporting goods store and you'll see licensed replicas of real guns. I don't expect there's a reliable way to tell if a good replica is a fake or not.
In a surprised %26amp; stressful situation, even people who are trained can't tell the difference.
Those pranksters may do that to the wrong person some day, and get the business end of the real deal.
